چکیده
SUMMARY Alkaline sucrose gradients were used to study the induc tion of single-strand breaks in and repair of rat liver DNA in vivo. Liver DNA was labeled during regeneration after partial hepatectomy. DNA was isolated from the organ in such a way as to minimize the induction of breaks due to handlingor enzymaticactivity.Differentmethodsof prepar ing high-molecular-weight liver DNA were compared. Best results were obtained by squashing the liver in an ethylene diaminetetraacetic acid-sodium chloride buffer. Using this squash technique and alkaline sucrose gradients, high-mo lecular-weight DNA was regularly obtained. This DNA be havedlike single-stranded DNA onhydroxyapatite columns and contained no detectable contamination of protein or RNA. It was demonstrated by the method described that hepatic DNA damage was produced within four hr by the administration of methyl methanesulfonateand that the rat can repair such hepatic DNA damage in vivo within 48 hr.

Studies on electron beam induced DNA damage and repair kinetics in lymphocytes by alkaline comet assay
Background: Exposure to ionizing radiation is known to induce oxidative stress followed by damage to critical biomolecules like lipids, proteins and DNA through radiolysis of cellular water. Since radiation has been widely used as an important tool in therapy of cancer, the detailed investigation regarding the DNA damage and repair kinetics would help to predict the radiation sensitivity of cel...
متن کاملOGG1 DNA Repair Gene Polymorphism As a Biomarker of Oxidative and Genotoxic DNA Damage
Background: Single nucleotide polymorphisms in 8-oxoguanine DNA glycosylase-1 (OGG1) gene modulates DNA repair capacity and functions as one of the first lines of protective mechanisms against 8-hydroxy-2’-deoxyguanosine (8-OHdG) mutagenicity. OGG1-Cys326 gene polymorphism may decrease DNA repair function, causing oxidative stress due to higher oxidative DNA damage. The main purpose of this stu...
متن کاملThein vitro and in vivo Effect of Clinoptiloliteon Decreasing of Copper Ion and DNA Damage of Anodonta Cygnea
The ability of natural zeolite (Clinoptilolite) to remove copper from aqueous ecosystem was studied in real and laboratory conditions. Fresh water mussels (Anodonta Cygnea) of different sizes were Exposed to copper (150, 350, 450 µgl-1) for 10 days. Copper exposure induced DNA damage in the haemolymph cells of Anodonta of all sizes. In connection with real samples, the amount of damage and even...
متن کاملRadioadaptive response in peripheral blood leukocytes of occupationally exposed medical staff with investigation of DNA damage by the use of neutral comet assay
Background: ˝Radioadaptive Response˝ is well-documented phenomenon appeared in low dose ionizing radiation received in vitro and in vivo. Occupational exposure has always been a great concern for radiation workers therefore this study was performed to study radioadaptive response in terms of residual DNA double strand breaks as an endpoint in peripheral blood leukocytes of occupationally expose...
متن کاملThe Role of Gene Therapy in Cartilage Repair
The key principle of gene delivery to articulations by direct intra-articular injection is to release complementary DNA(cDNA)-encoding medical products that will lead to maintained, endogenous production of the gene products withinthe articulation. In fact, this has been accomplished for both in vivo and ex vivo gene delivery, using several vectors, genes, and cells in some animal models. Some c...
